Stem-cell-derived human microglia transplanted into mouse brain to study human disease

چکیده

Microglia are critically involved in complex neurological disorders with a strong genetic component, such as Alzheimer’s disease, Parkinson’s disease and frontotemporal dementia. Although mouse microglia can recapitulate aspects of human physiology, they do not fully capture the reproduce all cell states. Primary cultures or derived from induced pluripotent stem cells (PSCs) difficult to maintain brain-relevant states vitro. Here we describe MIGRATE (microglia vitro generation refined for advanced transplantation experiments, which provides combined differentiation vivo xenotransplantation protocol study context brain. This article details an accurate, step-by-step workflow that includes PSCs, into brain quantitative analysis engraftment. Compared current protocols, present optimized, faster more efficient approach yields up 80% chimerism. To quantitatively assess engraftment efficiency by flow cytometry, access specialized cytometry is required. Alternatively, percentage chimerism be estimated standard immunohistochemical analysis. The takes ~40 d complete, culturing PSCs assessment. Mancuso De Strooper colleagues studying transplanted

Hallmarks of Alzheimer’s Disease in Stem-Cell-Derived Human Neurons Transplanted into Mouse Brain

Human pluripotent stem cells (PSCs) provide a unique entry to study species-specific aspects of human disorders such as Alzheimer's disease (AD). However, in vitro culture of neurons deprives them of their natural environment. Here we transplanted human PSC-derived cortical neuronal precursors into the brain of a murine AD model. Human neurons differentiate and integrate into the brain, express...

Fingolimod Enhances Oligodendrocyte Differentiation of Transplanted Human Induced Pluripotent Stem Cell-Derived Neural Progenitors

Multiple sclerosis (MS) is an autoimmune disease which affects myelin in the central nervous system (CNS) and leads to serious disability. Currently available treatments for MS mainly suppress the immune system. Regenerative medicine-based approaches attempt to increase myelin repair by targeting endogenous progenitors or transplanting stem cells or their derivatives. Fingolimod exerts anti-inf...

Purpose: Differentiation of mouse embryonic stem cells into Insulin secreting endocrine cells. Materials and Methods: In this study, Royan B1 mouse embryonic stem cell (derived from C57BL/6 mouse) were used. In directed differentiation method, embryonicstem cells after embryoid bodies formation were differentiated into insulin secreting cells. Nestin positive cells were obtained after culture ...
